HLA Bw4 and Bw6 Epitopes Recognized by Antibodies and …
To describe the structural basis of HLA Bw4 and Bw6 epitopes that are recognized by antibodies and the KIR3DL1 NK cell receptor. Molecular modeling and x-ray crystallography have refined our understanding of Bw4 and Bw6. These epitopes had been defined by comparison of HLA allele sequences and by site-directed mutagenesis.

Recent findings: Molecular modeling and X-ray crystallography have refined our understanding of Bw4 and Bw6. These epitopes had been defined by comparison of HLA allele sequences and by site-directed mutagenesis. Anti-Bw4 and anti-Bw6 antibodies and KIR3DL1 receptors recognize HLA α-1 α-helix residues 77-83 in combination with other HLA regions.
The Bw4 public epitope of HLA-B molecules confers reactivity …
These results show that the presence of the Bw4 epitope influences recognition of HLA-B molecules by NK cells that express NKB1, and suggest that the NKB1 molecule may act as a receptor for Bw4+ HLA-B alleles.
HLA-B*44 and the Bw4-80T motif are associated with poor …
HLA-B alleles are associated with outcomes in various pathologies, including autoimmune diseases and malignancies. The encoded HLA-B proteins are pivotal in antigen presentation to cytotoxic T cells, and some variants containing a Bw4 motif also serve as ligands to the killer immunoglobulin-like receptors (KIR) 3DL1/S1 of NK cells.
2023年11月21日 · Methods: We here combine the HLA allele frequencies in over six-million Hematopoietic Stem Cells (HSC) donors with a novel machine-learning-based method to predict allele frequency. Results: We show for the first time that allele frequency can be predicted from their sequences. This prediction yields a natural measure for selection.
Bw4-reactive and Bw6-reactive antibodies recognize multiple distinct ...
1994年11月1日 · To test whether antithetical structures are required to express Bw4 and Bw6 epitopes, we measured binding of Bw4-reactive and Bw6-reactive alloantibodies and mAbs to HLA-B7 variants. A triple substitution of HLA-B7 alpha-1 helix residues 80, 82, and 83 created Bw4 and destroyed Bw6 epitopes detected by alloantibodies and mAbs.

2012年10月1日 · Bw4 is an epitope present on several HLA-A and HLA-B antigens that can be divided into 8 subtypes based on amino acid differences (Tissue Ag 2000, 56:363). Recently, residues 80-83 have been shown to influence electrostatic potential of the Bw4 epitope, explaining differences in serological patterns (Human Immunol 2011, 72:1049).
Novel Human Anti-HLA-Bw4 and B61 Monoclonal Antibodies Kill …
2017年12月7日 · To overcome this problem, we recently created a novel method to develop mAbs using microarray technology and human peripheral blood (PB) B lymphocytes derived from donors who are positive for anti-HLA antibodies. The process takes approximately 1 month to produce human anti-HLA mAbs to treat myeloma and lymphoma.

2014年10月1日 · In our study, the presence of HLA-C1 and HLAC2 allotypes, HLA-B Bw4 and Bw6 dimorphism, as well as HLA-A Bw4 group, were detected using polymerase chain reaction with sequence-specific...
