A comparison of chemical shift sensitivity of trifluoromethyl tags ...
BTFA and TFET represent two commonly used 19 F NMR probes that are expected to minimally perturb target proteins due to their relatively small size. With the exception of TFET, all of the probes may be conjugated to free thiols in a single reaction step.
19F NMR as a tool in chemical biology - PMC
For example, 19 F NMR studies on cysteine-labelled ß2-adrenergic receptor (β2AR) using either a 19 F-BTFA [29] or a 19 F-TFET [36] tag were used to explore the differential long range conformational effects induced by agonists, inverse agonists, …
A comparison of chemical shift sensitivity of trifluoromethyl tags ...
The elucidation of distinct protein conformers or states by fluorine ( (19)F) NMR requires fluorinated moieties whose chemical shifts are most sensitive to subtle changes in the local dielectric and magnetic shielding environment. In this study we evaluate the effective chemical shift dispersion of a …
Current applications of 19F NMR to studies of protein structure …
2012年4月1日 · We will then address some of the common 19 F NMR approaches used in protein studies and the concomitant difficulties associated with assignment, line broadening, and labeling. Finally we highlight several recent results, which make use of these approaches in studies of protein folding, and ligand binding.
Analysis of the 19 F NMR spectra of BTFA-labeled cpSRP43
We attached the 3-bromo-1,1,1-trifluoropropan-2-one (BTFA) probe to single cysteines engineered at surface-exposed residues throughout cysteineless cpSRP43 and collected 1D 19 F-NMR spectra...
Monitoring conformational changes in the human …
2024年12月16日 · Dynamical data connecting the crystal structure to the transport cycle are critical to understanding how ligands are translated through the membrane. In the present study, we applied 19 F-based nuclear magnetic resonance (NMR) spectroscopy to 19 F labelled LeuT to monitor how substrates and ions binding affect the conformations of the transporter.
Defining a conformational ensemble that directs activation of PPARγ
A single well-populated 19 F NMR peak is observed for BTFA probes placed in two areas of the co-regulator-binding surface (helices 3 and 12) when PPARγ-BTFA is bound to a strong agonist such as GW1929 or rosiglitazone.
19F‐NMR Reveals the Role of Mobile Loops in Product and …
2017年3月3日 · Resistance to β‐lactam antibiotics mediated by metallo‐β‐lactamases (MBLs) is a growing problem. We describe the use of protein‐observe 19 F‐NMR (PrOF NMR) to study the dynamics of the São Paulo MBL (SPM‐1) from β‐lactam‐resistant Pseudomonas aeruginosa.
BTFA and TFET represent two commonly used 19F NMR probes that are expected to minimally perturb target proteins due to their relatively small size. With the exception of TFET, all of the probes may be conjugated to free thiols in a single reaction step.
A comparison of chemical shift sensitivity of trifluoromethyl tags ...
2015年3月27日 · To understand the origin of our experimental results, Density Functional Theory (DFT) calculations of the average 19 F NMR chemical shift were performed on three trifluoromethyl tags, anticipated to be most useful for labeling cysteine residues in NMR studies—namely, BTFA, TFET and BTFMA.
