Molecular mechanisms governing aquaporin relocalisation
2022年4月1日 · Drug discovery efforts towards pore-blocking AQP inhibitors have made little progress. AQPs are dynamically relocalised between intracellular vesicles and plasma membrane. This relocalisation represents a new target for inhibition of AQP function.
Aquaporin water channels in the nervous system - Nature
2013年3月13日 · Aquaporins (AQPs) are water channel proteins that increase cell membrane water permeability and assemble in cell membranes as tetramers. AQP4, the main water channel in the CNS, is...
Targeting Aquaporin-4 Subcellular Localization to Treat ... - Cell …
2020年5月14日 · Here we show that AQP4 cell-surface abundance increases in response to hypoxia-induced cell swelling in a calmodulin-dependent manner. Calmodulin directly binds the AQP4 carboxyl terminus, causing a specific conformational …
Aquaporins: Current Biology - Cell Press
2013年1月21日 · Data from cell and animal models provide a compelling rationale for the development of aquaporin modulators as therapeutics: AQP1 inhibitors are predicted to have utility in diuretic-refractory edema, tumor angiogenesis and glaucoma; AQP4 inhibitors in some forms of brain edema and injury; and AQP3 inhibitors in some cancers.
Aquaporins implicated in the cell proliferation and the signaling ...
2021年9月1日 · Regulation of osmotic homeostasis via AQPs is accompanied by dynamic participation of various cellular signaling pathways. Recently emerging evidence reveals that functional roles of AQPs are further extended from the osmotic regulation via water permeation into the cell proliferation and differentiation.
Aquaporins and cell migration - PMC
Aquaporin (AQP) water channels are expressed primarily in cell plasma membranes. In this paper, we review recent evidence that AQPs facilitate cell migration. AQP-dependent cell migration has been found in a variety of cell types in vitro and in mice in vivo. AQP1 deletion reduces endothelial cell migration, limiting tumor angiogenesis and growth.
Rapid Aquaporin Translocation Regulates Cellular Water Flow
2012年3月30日 · In this study, we combined laser scanning confocal microscopy of chimeras of AQP1 with green fluorescent protein (AQP1-GFP), calcium imaging, and mutagenesis to determine that AQP1 translocation underpins the regulation of cellular water flow, as measured by changes in cell volume.
Immunohistochemical Localization of the Aquaporins AQP1, …
Some cell types, such as renal collecting duct cells, also permeate a larger quantity of water and in these cases the membrane water channel known as aquaporin (AQP) serves as the molecular machinery for such rapid and efficient transfer through the membrane, as first described by Agre and colleagues in 1992 .
Unprecedented Cell-Selection Using Ultra-Quick Freezing …
2014年2月18日 · Here we show that membrane damage caused by ultra-quick freezing is rescued by the expression of AQP4. We next examine if the expression of AQP combined with ultra-quick freezing can be used to select cells efficiently under freezing conditions where most cells are died.
