2024年1月12日 · We report for the first time the combination of directed evolution focused on enhancing and reversing the stereoselectivity of an enzyme with Cu(I)-mediated click chemistry (CuAAC), providing an asymmetric click approach for versatile chiral triazoles products.

Among these reactions, the cycloaddition of cyclopropenes and their analogues to ortho -quinones has been established as a highly promising click reaction. The present work investigates the mechanism involved in the cycloaddition of strained dienes to ortho -quinones and structural factors that would influence this reaction.

In this review, we discuss the application of click chemistry in peptide-based drug design. We highlight how triazoles formed by click reactions have been used for mimicking peptide and disulfide bonds, building secondary structural components of peptides, linking functional groups together, and bioconjugation.
