Our protocol describes the specific steps for analyzing dsRNA binding and cleaving by Dicer protein in PaKiT03 cells, after transfecting a pre-synthesized dsRNA. In our study, dsRNA was cleaved into small RNAs by Dicer, thus we applied a small RNA process pipeline to identify dsRNA-derived small RNAs.

DICER function dsRNA and pre-miRNA processing. DICER was firstly recognized for its role in siRNA processing. In a seminal work, Bernstein and colleagues showed that D. melanogaster DICER cleaves dsRNA precursors into ~ 22-nt small dsRNAs revealing a fundamental role for DICER in the RNAi pathway (Bernstein et al. 2001).
