DIO2 solely catalyzes thyroid hormone activation. It is expressed in brain, brown adipose tissue, bone, muscles, cochlear, pituitary, etc. An impaired expression of DIO2 might manifest in humans as mental retardation, probably combined with impaired hearing (Campos-Barros et al., 2000).

Once T 4 enters a target cell (primarily in the thyroid or brain), it is converted rapidly to T 3 by cytoplasmic type 2 deiodinase (DIO2). DIO2 produces most T 3 in the human body, about 20 μg per day. DIO1 produces a much smaller amount of T 3, roughly 5 μg per day.
