2025年2月12日 · LC3, a central autophagy protein, exists in lipidated (LC3-II) and non-lipidated (LC3-I) forms, both critical for neurons due to their sensitivity to metabolic and proteostatic stress. To elucidate the specific roles of membrane-anchored LC3A/B in post-mitotic neurons, we engineered deconjugases with enhanced selectivity for lipidated LC3.

Omrane and Ben M’Barek et al. found that long-term starvation triggers a noncanonical autophagic process on lipid droplets (LDs). This process involves the binding of Atg3 to specific, ultra-large micrometric LDs, lipidation of LC3B to these LDs, and recruitment of acidified autophagosome-like membranes for degradation.

Time-lapse observations suggested the close association between LC3 surge in a neurite and its specification toward an axon. Trehalose and bafilomycin accelerated and delayed axonal specification respectively in a developing neuron.

The LC3B-II/LC3B-I ratio, an index of autophagosome formation, was lower in the neurons derived from the AD patient with APP-E693Δ as well as the neurons derived from other familial and sporadic AD patients. These findings indicate that dysregulation of autophagy-related molecules may accelerate the pathogenesis of AD.
