LGR5 increases the affinity of RSPO1 to RNF43 without contacting RNF43. This study reveals the structural basis of these interactions.

LGR5 is a membrane protein expressed in many normal cells and tumor cells. The binding of LGR5 to its ligand R-spodin enhances the WNT-beta-catenin signaling pathway and leads to tumor progression. LGR5 may play a key role in the maintenance of stem cell properties, and can serve as a marker for cancer stem cells.
