The role of MukEF is unclear. MukEF disrupts MukB-DNA interactions in vitro. In vivo, however, MukEF stimulates MukB-induced DNA condensation and is required for the assembly of MukB clusters at the quarter positions of the cell length. We report here that MukEF is essential for stable association of MukB with the chromosome.
Structural Studies of a Bacterial Condensin Complex Reveal ATP ...
2009年1月9日 · We report complete structural views of the intersubunit interactions of this condensin along with ensuing studies that reveal a role for the ATPase activity of MukB. MukE and MukF together form an elongated dimeric frame, and MukF's C-terminal winged-helix domains (C-WHDs) bind MukB heads to constitute closed ring-like structures.
The Role of MukE in Assembling a Functional MukBEF Complex
2011年9月30日 · In a manner similar to SMC, MukB requires the association of two accessory proteins (MukE and MukF) for its function. MukF is a constitutive dimer that bridges the interaction between MukB and MukE. While MukB can condense DNA on its own, it requires MukF and MukE to ensure proper chromosome segregation.
Chromosome Condensation in the Absence of the Non-SMC …
MukBEF is a bacterial SMC (structural maintenance of chromosome) complex required for chromosome partitioning in Escherichia coli. We report that overproduction of MukBEF results in marked chromosome condensation. This condensation is rapid and precedes the effects of overproduction on macromolecular synthesis.
Dynamic architecture of the - Oxford Academic
2019年8月10日 · By analysing changes in the architecture and stoichiometry of the Escherichia coli SMC complex, MukBEF, as a function of nucleotide binding to MukB and subsequent ATP hydrolysis, we demonstrate directly the formation of dimer of MukBEF dimer complexes, dependent on dimeric MukF kleisin.
Cells lacking func-tional MukBEF, grown in a minimal medium under permissive conditions, have disorganized chromosomes with misplaced genetic loci, and fail to properly segregate sister...
DNA sliding and loop formation by E. coli SMC complex: MukBEF
2022年9月1日 · MukEF depleted strains show the same temperature-sensitive growth phenotype as the MukB-null strain, and MukEF is essential for stable association of MukB with chromosome in vivo [37].
MukBEF, a chromosomal organizer - PMC
MukBEF acts as a complex molecular machine that assists in chromosome segregation and global organization. The review focuses on mechanistic analysis of DNA organization by MukBEF with the emphasis on its involvement in formation of chromatin scaffold and plausible other roles in chromosome segregation.
Towards the architecture of the chromosomal architects
MukBEF, the bacterial prototype of eukaryotic condensins and cohesins, has a key role in the global chromosomal organization of Escherichia coli and its close relatives.
A folded conformation of MukBEF and cohesin - PMC
Using structural biology, mass spectrometry and cross-linking, we investigated the architecture of two evolutionarily distant SMC–kleisin complexes: MukBEF from Escherichia coli and cohesin from Saccharomyces cerevisiae.
