Nephrin is a 180 KD trans-membrane protein expressed in glomerular podocytes. It was first identified in children with congenital nephrotic syndrome of the Finnish type (NPHS1).

Sep 1, 2007 · Nephrin is a transmembrane protein (∼185–200 kDa) with a short intracellular domain, and an extracellular domain with eight distal IgG-like motifs and one proximal fibronectin type III-like motif (Figure 2 a). The length of the entire extracellular domain of nephrin is ∼35 nm.

Nephrin is a key molecule in podocytes to maintain normal slit diaphragm structure. Nephin interacts with many other podocyte and slit diaphragm protein and also mediates important cell signaling pathways in podocytes.

Nephrin Phosphorylation Establishes a Signaling Platform Beyond its role as a physical barrier, nephrin acts a central signaling platform within the podocyte, facilitated by an extensive network of cytoplasmic binding partners which have been identified over the past two decades (Figure 1 B) (18, 29, 45 – 76).

Nephrin is a key podocyte protein, which regulates the actin cytoskeleton via tyrosine phosphorylation of its cytoplasmic domain. Here we report that two protein tyrosine phosphatases, PTP1B and PTP-PEST negatively regulate nephrin tyrosine phosphorylation.

Nephrin is a unique molecular component of the podocyte slit diaphragm. We have learned a lot about nephrin and its role in the renal ultrafilter, but there are still several important questions that remain to be answered (Box 1).

Nephrin is a central component of the glomerular ultrafilter, with both structural and signaling functions. The extracellular domain of nephrin and other components of the slit diaphragm seem to form a porous molecular sieve.

Jul 7, 1999 · In the present study, we localized nephrin in immunofluorescence microscopy to the GBM region of newborn human glomeruli, using antibodies generated against recombinant antigen. Moreover, we demonstrated by immunoelectron microscopy that nephrin is located in the podocyte slit region.
