SIRT1 and p53, effect on cancer, senescence and beyond
There is new evidence that SIRT1 acts as a tumor suppressor based on its role in negatively regulating beta-catenin and survivin. This review provides an overview of current knowledge of SIRT1-p53 signaling and controversies regarding the functions of SIRT1 in tumorigenesis.

Sirtuin1-p53: A potential axis for cancer therapy - PubMed
P53 is one of the most studied tumor suppressors and the first identified non-histone target of SIRT1. SIRT1 deacetylates p53 in a NAD +-dependent manner and inhibits its transcriptional activity, thus exerting action on a series of pathways related to tissue homeostasis and various pathological states. The SIRT1-p53 axis is thought to play a ...
SIRT1 and p53, effect on cancer, senescence and beyond
Several studies support the notion that SIRT1 could be involved in carcinogenesis: first, SIRT1 knockout mice exhibit p53 hyperacetylation and increased radiation-induced apoptosis consistent with SIRT1 inhibition of p53 function, raising the possibility that SIRT1 can facilitate tumor growth by antagonism of p53 ; second, inhibition of SIRT1 ...
P53 and Sirt1: Routes of metabolism and genome stability
2014年11月1日 · SIRT1 modulates both p53 transcription-dependent and p53-independent apoptosis. The latter is initiated through cytochrome c release from the mitochondria. Emerging evidence indicates that SIRT1 prevents p53 nuclear translocation.
Inhibition of SIRT1 deacetylase and p53 activation uncouples
2019年2月11日 · We demonstrate these NSAIDs counteract carcinogen-induced proliferation by inhibiting the sirtuin 1 (SIRT1) deacetylase activity, augmenting acetylation and activity of the tumour suppressor...
Role of Sirtuin1-p53 regulatory axis in aging, cancer and cellular ...
SIRT1-p53 pathway regulates calorie restriction, longevity, cell cycle progression, inflammation and mitochondrial biogenesis/function in aging while telomere elongation, tumourigenesis, DNA repair, cell death/apoptosis and cell cycle arrest are the regulated aspects implicated in cancer.
Role of Sirtuin1-p53 regulatory axis in aging, cancer and cellular ...
SIRT1 deacetylates p53 in a NAD+-dependent manner to inhibit transcription activity of p53, in turn modulate pathways that are implicated in regulation of tissue homoeostasis and many disease states. In this review, we discuss the role of SIRT1-p53 pathway and its regulatory axis in the cellular events which are implicated in cellular aging ...
SIRT1: Regulator of p53 Deacetylation - PMC
The significance of SIRT1 deacetylation on p53, a tumor suppressor that is mutated in a large number of tumors, suggests that the SIRT1-p53 axis is a targetable pathway to regulate p53 tumor suppression activity . Over the years, many endogenous factors and small molecules have been identified that can directly affect the SIRT1-p53 pathway.
SIRT1 is an actionable target to restore p53 function in HPV
2023年10月14日 · We show that SIRT1 inhibition restores a transcriptionally active K382-acetylated p53 in HPV + but not HPV - cell lines, which in turn promotes G 0 /G 1 cell cycle arrest and inhibits...
