SIRT6 links histone H3 lysine 9 deacetylation to NF-kappaB ... - PubMed
2009年1月9日 · SIRT6 interacts with the NF-kappaB RELA subunit and deacetylates histone H3 lysine 9 (H3K9) at NF-kappaB target gene promoters. In SIRT6-deficient cells, hyperacetylation of H3K9 at these target promoters is associated with increased RELA promoter occupancy and enhanced NF-kappaB-dependent modulation of gene expression, apoptosis, and cellular ...
SIRT6 inhibits inflammatory response through regulation of …
The present study showed that TNF-α significantly reduced the expression of SIRT6 protein and mRNA in a concentration- and time-dependent manner and increased the expression of monocyte chemotactic protein 1 (MCP-1), interleukin (IL) -6 and IL-1β in human umbilical vein endothelial cells (HUVECs).
Sirt6 mediates antioxidative functions by increasing Nrf2 abundance
2023年1月1日 · Sirt6, a member of the mammalian homolog family of yeast Sir2 NAD + -dependent histone deacetylases, regulates multiple biological processes. Sirt6 exerts antioxidative functions by enhancing DNA repair and DNA end resection. In our study, we found that Sirt6 expression was induced by H 2 O 2 and paraquat (PQ) in cells.
Structure and Biochemical Functions of SIRT6 - ScienceDirect
2011年4月22日 · SIRT6 is a member of the evolutionarily conserved sirtuin family of NAD +-dependent protein deacetylases and functions in genomic stability and transcriptional control of glucose metabolism. Early reports suggested that SIRT6 performs ADP-ribosylation, whereas more recent studies have suggested that SIRT6 functions mainly as a histone deacetylase.
SIRT6: Novel Mechanisms and Links to Aging and Disease
SIRT6, a member of the Sirtuin family of NAD +-dependent enzymes, has established roles in chromatin signaling and genome maintenance. Through these functions, SIRT6 protects against aging-associated pathologies including metabolic disease and …
SIRT6 inhibits inflammatory response through regulation of NRF2 …
2021年10月1日 · We found that SIRT6 promoted NRF2 phosphorylation. SIRT6 overexpression increased mRNA levels of NRF2 target genes and SIRT6 knockdown decreased mRNA levels of NRF2 target genes. Therefore, SIRT6 positively regulated phosphorylation of NRF2 to stimulate the transcription of its target genes to inhibit inflammation in endothelial cells.
Sirt6 attenuates chondrocyte senescence and osteoarthritis
2022年12月10日 · Here we show that Sirt6 deficiency exaggerates chondrocyte senescence and osteoarthritis progression, whereas intra-articular injection of adenovirus-Sirt6 markedly attenuates surgical...
SIRT6 is a key regulator of mitochondrial function in the brain
2023年1月18日 · Our analysis reveals that SIRT6 is a central regulator of mitochondrial activity in the brain. SIRT6 deficiency in the brain leads to mitochondrial deficiency with a global downregulation of...
SIRT6 safeguards human mesenchymal stem cells from oxidative …
2016年1月15日 · Our study uncovers a novel function of SIRT6 in maintaining hMSC homeostasis by serving as a NRF2 coactivator, which represents a new layer of regulation of oxidative stress-associated stem cell...
Structural Basis of Sirtuin 6-Catalyzed Nucleosome Deacetylation
2023年3月17日 · The sirtuins are a family of NAD-dependent HDAC enzymes, and one member, sirtuin 6 (Sirt6), influences DNA repair, transcription, and aging. Here, we demonstrate that Sirt6 is efficient at deacetylating several histone H3 acetylation sites, including its canonical site Lys9, in the context of nucleosomes but not free acetylated histone H3 ...
