SOFAST-HMQC experiments for recording two-dimensional ... - PubMed
Guidelines are provided on how to set up SOFAST-HMQC experiments for a given protein sample. In addition, an alternative pulse scheme, IPAP-SOFAST-HMQC is presented that allows application on NMR spectrometers equipped with cryogenic probes, and fast measurement of one-bond 1H-13C and 1H-15N scalar and residual dipolar coupling constants.

2020年8月3日 · Here, we present the XL-ALSOFAST- [13 C, 1 H]-HMQC experiment with much improved sensitivity for fully protonated high molecular weight proteins. For the tested systems ranging from 100 to 240 kDa in size, 3-fold higher sensitivity was obtained on average for fast relaxing signals compared to current state-of-the-art experiments.

Go to an existing SOFAST experiment (pulse program: sofast), type “edc” to copy its parameters and generate a new experiment number. Or "rpar SFHMQCF3GPPH" in an new experiment entry. Type “ ased ” to adjust power length and power level for both 1H and 15N, including, pL1, p3, pL2, pL12, and pcpd2.
After the pulse z-magnetization is recovering with a typical rate, the T1-relaxation time. This magnetization is used for the next pulse and the relation between the pulse angle α and the ratio T/T1 is important α. A.Ross, M. Salzmann, H. Senn J. Biomol. NMR 10, 389-396 (1997) 1 has an influence on the repetition rate.
SOFAST HMQC - SpringerLink
The main features of the SOFAST HMQC experiment are (1) the use of shaped 1 H pulses that manipulate only a subset of proton spins in order to enhance their spin–lattice relaxation (T 1) via dipolar interactions with the unperturbed proton spins; and (2) Ernst angle excitation to achieve maximal sensitivity for high repetition rates of the pulse...
