TRECS - Overview: T-Cell Receptor Excision Circles Analysis, Blood
The genomic DNA and T-cell receptor excision circles (TREC) in the cell lysates are quantified in the real-time PCR assay, in triplicate, by using a fluorescent probe specific for the T-cell receptor delta-deletion TREC signal joint and a distinct fluorescent probe for the reference gene, albumin.
TREC Based Newborn Screening for Severe Combined Immunodeficiency ...
Newborn screening (NBS) by quantifying T cell receptor excision circles (TRECs) in neonatal dried blood spots (DBS) enables early diagnosis of severe combined immunodeficiency disease (SCID). In recent years, different screening algorithms for …
T-cell receptor excision circles - Wikipedia
T-cell receptor excision circles (TRECs) are small circles of DNA created in T-cells during their passage through the thymus as they rearrange their TCR genes. [1] . Their presence indicates maturation of T cells; TRECs are reduced in SCID (severe combined immunodeficiency disease).
Newborn Screening for Severe Combined Immunodeficiency
The T-cell receptor excision circle (TREC) assay is an effective screening tool for severe combined immunodeficiency (SCID). The TREC assay was designed to detect typical SCID and leaky SCID, but any condition causing low naïve T-cell counts will also be detected.
Newborn screening using TREC/KREC assay for severe T and B cell ...
2018年6月26日 · By measuring the copy numbers of T cell receptor excision circles (TRECs) and κ-deleting excision circles (KRECs), it is possible to identify T and B lymphopenia related disorders (including SCID, X-linked agammaglobulinemia (XLA) and ataxia telangiectasia (AT)).
A Practical Approach to Newborn Screening for Severe Combined ...
Severe combined immunodeficiency (SCID) is a life-threatening condition of newborns and infants caused by defects in genes involved in T cell development. Newborn screening (NBS) for SCID using the T cell receptor excision circle (TREC) assay began ...
Newborn Screening for Severe Combined Immunodeficiency-A History …
2017年6月18日 · The TREC assay, initially developed by Douek et al. to measure changes in thymic output following HIV infection and treatment, has been optimized for use in NBS for T cell lymphopenias and SCID, being both sensitive and specific.
Newborn Screening through TREC, TREC/KREC System for Primary ...
Global newborn screening for PID, using an assay to detect T-cell receptor excision circles (TREC) in dried blood spots (DBS), is now being performed in all states in the United States. In this review, we discuss the development and outcomes of TREC, TREC/KREC combines screening, and continued challenges to implementation.
Establishing Simultaneous T Cell Receptor Excision Circles (TREC) and …
We have shown a good correlation between the TREC results using our established assay with the CDC in-house method, indicating this assay is potentially applicable for the NBS program for severe PIDD. Our TREC and KREC assay was able to correctly identify the 11 selected PIDD patients with aberrant T and/or B cell counts.
2022年7月12日 · Test Indications: This assay is useful for evaluating thymic function in patients with cellular or combined primary immunodeficiencies, or receiving immunotherapy or cancer vaccines.
