U.S. Route 521 (US 521) is a north–south United States Highway that traverses 177.3 miles (285.3 km), from Georgetown, South Carolina, to Charlotte, North Carolina.

U.S. Route 521 Truck (US 521 Truck) is a truck route of US 521 to direct truck traffic to avoid downtown Camden. The highway takes various roads including: Ehrenclou Drive, Chestnut Ferry Road, Jefferson Davis Highway (in concurrency with US 1 / US 601 / SC 34), Springdale Drive, and Boykin Road (in concurrency with US 601 Truck).

The NCDOT has plans to widen US 521 between Brixham Hill Ave. just south of I-485 and the South Carolina border in Mecklenburg County. Locally known as Johnston Road or the Lancaster Highway, depending on location, it sees heavy traffic daily thanks to the growth and popularity of the Ballantyne area in south Charlotte.
