NK cells expressing dominant-negative Rac1 have a decreased ability to form conjugates with targets, and those that do form conjugates have decreased ability to polarize their cytolytic granules toward the target cell. Together, our data highlight a regulatory role for Vav and Rac1 in the generation of cell-mediated killing.

1999年3月15日 · In this report we have addressed the role of the specific Rac1 activator, Vav, in the generation of NK cell cytotoxic activity. Our findings show that the activation of NK cells through FcγRIII or by interaction with sensitive target cells results in the rapid stimulation of Vav tyrosine phosphorylation.

Our data provide the first evidence to implicate VAV2 in glucose-induced Rac1 activation, actin remodelling and GSIS in pancreatic beta cells.
