VHH-Based Bispecific Antibodies Targeting Cytokine Production
These bispecific antibodies combine an anti-TNF V H H with V H Hs or scFvs directed against abundant surface molecules on myeloid cells and serve to limit the bioavailability of TNF produced by these cells. Such reagents may become prototypes of a novel class of anti-cytokine biologics.

2024年10月18日 · The VHH NGS data from Deszynski et al. (2022) holds extensive information regarding sequence variability, dependencies and relationships between amino acids at specific positions, potentially reflecting functional and structural relevance.
General Strategy to Humanize a Camelid Single-domain Antibody …
These immunoglobulins are referred to as heavy-chain antibodies (1), and their antigen-binding fragment is reduced to a single domain (referred to as VHH or Nanobody), with a molecular size of only ∼15 kDa, which is smaller in comparison to single-chain variable fragment fragments (30 kDa), Fab fragments (60 kDa), and whole antibodies (150 kDa).
An alpaca single-domain antibody (VHH) phage display library ...
2022年7月26日 · Antigen-combining sites of the camelid heavy-chain antibody variable domain (VHH) are constructed by three complementarity-determining regions (CDR1, CDR2 and CDR3). We prepared cDNA using mRNA extracted from peripheral lymphocytes of alpacas that had been non-immunized or immunized with human serum …
Nanobody, or VHH, is the variable segment of camel heavy chain...
Heavy‐chain antibodies (HCAbs) are a unique type of antibodies devoid of light chains, and comprised of two heavy chains‐only that recognize their cognate antigen by virtue of a single...
Construction of a Humanized Artificial VHH Library Reproducing ...
A variable domain of heavy chain antibody (VHH) has different binding properties than conventional antibodies. Conventional antibodies prefer binding to the convex portion of the antigen, whereas VHHs prefer epitopes, such as crevices and clefts on ...
Molecular recognition requires dimerization of a VHH antibody
Camelid heavy-chain-only antibodies are a unique class of antibody that possesses only a single variable domain (termed VHH) for antigen recognition. Despite their apparent canonical mechanism of target recognition, where a single VHH domain binds a single target, an anti-caffeine VHH has been observed to possess 2:1 stoichiometry.
Characterization of heavy-chain antibody gene repertoires in …
2023年1月1日 · To characterize the diversity of variable genes of HCAbs (VHHs), germline and rearranged VHH repertoires are constructed. Phylogenetics analysis shows that all camelid VHH genes are derived from a common ancestor and the nucleotide diversity of VHHs is similar across all camelid species.
