Transcription of rDNA in the Yeast Saccharomyces cerevisiae
In the yeast S. cerevisiae, approximately 150 rDNA repeats are tandemly arranged on chromosome XII. A single unit of rDNA contains two transcribed genes (fig. 1A), the 35S rRNA gene that is transcribed by Pol I and the gene for 5S rRNA, which is transcribed by RNA polymerase III (Pol III).
High plasticity of ribosomal DNA organization in budding yeast
2024年2月27日 · Our results indicate that rDNA in budding yeast tolerates radical changes in organization and provide tools for rDNA engineering in synthetic genomes. To engineer the rDNA organization, we need to have the capacity to efficiently build rDNA arrays with designed sequences at chosen target sites.
The yeast rDNA locus: A model system to study DNA
2005年7月28日 · The yeast rDNA locus is a good model to study DNA repair in chromatin for the following reasons: (1) removal of CPDs is very efficient, (2) it occurs via both pathways of NER, (3) the rDNA chromatin is well characterized, and (4) ribosomal genes are partitioned between heterochromatin and euchromatin.
rDNA array length is a major determinant of replicative lifespan in ...
Here, we show that variation in copy number at the rDNA locus is capable of altering replicative lifespan in yeast. These results indicate that considering rDNA copy number, and conditions that could potentially change this dynamic chromosome locus, is critical for evaluating lifespan.
Cellular Senescence in Yeast Is Regulated by rDNA Noncoding ...
2013年9月23日 · Studies in the yeast Saccharomyces cerevisiae over the past 20 years have found a connection between the ribosomal RNA gene cluster (rDNA) and lifespan [2]. The highly repetitive rDNA exhibits genomic instability, and the antiaging histone deacetylase gene SIR2 regulates this instability.
Nucleolar and Ribosomal DNA Structure under Stress: Yeast …
Simple model organisms like the budding yeast Saccharomyces cerevisiae, easy to manipulate genetically, are useful in order to study nucleolar and rDNA structure and their relationship with stress. In this review, we summarize the most important findings related to this topic.
RNA polymerase I in yeast transcribes dynamic nucleosomal rDNA
2007年1月28日 · Using a yeast strain with reduced numbers of all actively transcribed rDNA repeats, we show that rDNA exists in a dynamic chromatin structure of unphased nucleosomes. Furthermore, it is...
Nucleolar structure: Yeast rDNA feels the heat - PMC
2017年7月19日 · Using this sensitive method, the authors make the unexpected discovery that rDNA structure is highly sensitive to temperature in yeast cells; yeast allowed to progress into mitosis at the elevated temperature of 37C adopt a novel rDNA morphology that they interpret as a hypercondensed state.

2023年12月21日 · The molecular requirements for H3K9 methylation within the rDNA locus in fission yeast are distinct from those in other heterochromatin domains. Several key components of the conventional RNAi pathway, such as the Clr4 histone methyltransferase and the argonaute protein Ago1, are essential for effecting H3K9 methylation at the rDNA site (Hirai ...
Regulators of rDNA array morphology in fission yeast
2024年7月5日 · We surveyed hundreds of yeast mutants to identify factors that regulate rDNA spatial organization, discovering that the deletion of ribosome protein genes causes rDNA array expansion. Further experiments indicated that ribosome protein gene insufficiency activates conserved regulatory signaling pathways as part of a compensatory response.
