Atg17 regulates the magnitude of the autophagic response
The atg17delta mutant forms a reduced number of small autophagosomes. As a result, it is defective in peroxisome degradation and is partially defective for autophagy. Atg17 interacts with both Atg1 and Atg13, via two coiled-coil domains, and these interactions facilitate its inclusion in the Atg1 complex.

2012年12月21日 · ATG17-GFP cells display bright perivacuolar puncta, corresponding to the PAS in both SMD (SMD, synthetic minimal medium containing 0.67% yeast nitrogen base with ammonium sulfate, 2% glucose, and supplemented with the appropriate amino acids for selection) and SMD with rapamycin treatment (Figure 5A).

Here we characterized Atg17/FIP200, the Drosophila ortholog of mammalian RB1CC1/FIP200, a proposed functional equivalent of yeast Atg17. Atg17 disruption inhibits basal, starvation-induced and developmental autophagy, and interferes with the programmed elimination of larval salivary glands and midgut during metamorphosis.

Atg17 Functions in Cooperation with Atg1 and Atg13 in Yeast …
ATG17 was first identified by a two-hybrid screen using ATG1 as bait (Kamada et al., 2000). ATG17 is required specifically for autophagy, but its precise role remains to be determined. The fact that overexpression of Atg1 rescues the autophagic defect in atg17 Δ cells suggests that Atg17 acts upstream of Atg1.
Atg17 Regulates the Magnitude of the Autophagic Response
An N-terminal GFP fusion to full-length Atg17 restored the Pho8Δ60 activity of the atg17Δ strain to ∼67% that of the wild-type strain, indicating that this construct retained substantial Atg17 function .

2008年4月25日 · Approximately 30 autophagy-related (Atg) proteins have been identified in S. cerevisiae, 17 of which are essential for formation of the autophagosome (reviewed in 17). Null mutations in most of these genes prevent induction of autophagy, and cells do not survive nutrient starvation; however, these mutants are viable in rich medium.

2016年1月1日 · The Atg17-Atg31-Atg29 Complex Interacts with the Vam7 SNARE Protein at the PAS. The Atg17-Atg31-Atg29 complex is a scaffold that is vital for PAS organization during autophagy induction. In a genome-wide study of protein-protein interaction via the yeast two-hybrid (Y2H) assay, Atg17 was found to bind Vam7, a vacuolar SNARE protein . This ...

2012年12月21日 · Atg17 is crescent shaped with a 10 nm radius of curvature. Dimerization of the Atg17-Atg31-Atg29 complex is critical for both PAS formation and autophagy, and each dimer contains two separate and complete crescents. Upon induction of autophagy, Atg17-Atg31-Atg29 assembles with Atg1 and Atg13, which in turn initiates the formation of the phagophore.
Atg17 functions in cooperation with Atg1 and Atg13 in yeast
Two-hybrid analyses and coimmunoprecipitation experiments demonstrated that Atg17 physically associates with Atg1-Atg13 complex, and this binding was enhanced under starvation conditions. Atg17-Atg1 binding was not detected in atg13delta mutant cells, suggesting that Atg17 interacts with Atg1 through Atg13.
