CRISPR-Screen又称全基因组CRISPR，或者高通量基因编辑技术，是一种由张锋团队开发的，基于 CRISPR-Cas9系统 ，通过构建sgRNA敲除文库同时对多个活性细胞的不同基因进行基因敲除，辅助以特定的筛选方案，通过 NGS测序 和生信分析的手段，预测出符合条件的候选基因。

CRISPR-Cas9技术直接靶向基因组、形成 indel （insertion or deletion），造成 基因移码突变 （基因沉默）。 与传统的 RNAi （RNA interference）相比，Cas9最大的优势在于更低的脱靶率。 基于Cas9诸多优势，全球各个实验室利用Cas9技术在多种细胞系及各种模式动物上进行实验，成果显著。 然而，单纯的设计一条或多条sgRNA（single guide RNA）进行基因敲低，这样的实验方法通量过低，因此，借鉴shRNA Screen技术，许多团队开发出版本各异的Cas9 Screen技术。
