Efficient production of recombinant human transcription factor IIE
2004年4月1日 · In this paper, we show that our recombinant human TFIIE is fully functional to stabilize a pre-initiation complex, consisting of promoter DNA, TBP, TFIIB, RNA polymerase II, and TFIIF. Our recombinant TFIIE also supports TFIIH-dependent and associated ATP-dependent activities in initiation and promoter escape.

We found that creating a region of heteroduplex DNA around the start site of the adenovirus major late (AdML) promoter circumvents the requirement for TFIIE and TFIIH in transcription.
We propose that IIH functions as a molecular wrench: rotating downstream DNA relative to Þxed upstream protein-DNA interactions, thereby generating torque on, and melting, the intervening DNA.
