Recently a class of NLSs known as PY-NLSs has been proposed, originally by Lee et al.[10] This PY-NLS motif, so named because of the proline - tyrosine amino acid pairing in it, allows the protein to bind to Importin β2 (also known as transportin or karyopherin β2), which then translocates the cargo protein into the nucleus.

In this paper, we propose a novel algorithm for NLS prediction based on sequential-pattern mining and linear motif scoring. Our strategy is first to detect potential NLS candidates using the sequential-pattern mining method, which are then scored in terms of their likelihood of being (part of) NLS based on their sequence and linear motif features.

Available data strongly suggest that simple karyophilic clusters of arginines and lysines in nucleus-targeted proteins signal the anchoring of these proteins to specialized transporter molecules found on the pore complex or in the cytoplasm. These peptides have been termed nuclear localization signals (NLS).

A database of NLS motifs is therefore a useful resource for biologists in identifying targeting signals in their sequence. The database describes all experimentally determined NLS motifs with links to original references in PubMed ( 7 ).

In recent years, NLS were widely used in cancer treatment and viral infection prevention [13–15], and researchers paid more attention to identifying novel NLS motifs and the import nucleoporins that recognize and bind them. Here, we categorized NLS according to their residue composition into classical (cNLS), non-classical (ncNLS), and other types.
