Here, we summarize recently published research on the NS5 protein, an important enzyme drug target. NS5 is the largest (102 kDa) and the most conserved protein (with ~70% sequence identity among the four serotypes, see Figure 1A) expressed during infection by dengue virus.

In this review, we discuss the functions of NS5, diverse NS5-mediated strategies adopted by flaviviruses to evade the host antiviral response, and how NS5 can be a target for the development of vaccines and antiviral therapeutics.
