Pennsylvania Route 340 (PA 340) is a 30-mile-long (48 km) state highway located in Lancaster and Chester counties in Pennsylvania. The western terminus is at PA 462 in Lancaster. The eastern terminus is at U.S. Route 30 Business (US 30 Bus.) in Thorndale.

The state route follows Old Philadelphia Pike through Lancaster County and Kings Highway through most of Chester County. Schedule time to visit when you get to Lancaster and it will offer some great scenery. The road has a lot of straightaways and twisties.

U.S. Route 340 (US 340) is a spur route of US 40, and runs from Greenville, Virginia, to Frederick, Maryland. In Virginia, it runs north–south, parallel and east of US 11, from US 11 north of Greenville via Waynesboro, Grottoes, Elkton, Luray, Front Royal, and Berryville to the West Virginia state line.

US 30 Eastbound Ramps intersect PA Route 340 at a stop-controlled intersection. Existing traffic signal remains at the intersection of PA Route 340 and Humpton Road/Turkey Hill. West Bondsville Road remains open. US 30 Mainline has three lanes in each direction.

The existing interchange of US 30 and PA 340 provides both eastbound and westbound on-ramps and off-ramps. Some of the significant environmental features of the section include, potential historic properties, a golf club, seven streams, eleven wetlands, and township open space.
