Parabrachial neuron activity is enhanced during chronic pain, and inactivation of PBN neurons in mice prevents the establishment of neuropathic, chronic pain symptoms.

With independent demonstration of LH PBN acute pain responses and the effect of these neurons on feeding motivation, we hypothesized that acute nociception may acquire LH PBN activity to suppress feeding.

The PBN is comprised of distinct pain pathways that play an important role in the manifestation of emotional behavior, autonomic reaction, and learning due to pain, especially in animals.

Hyperactivity of neurons in the parabrachial nucleus (PBN) is causally linked to exaggerated pain behaviors in rodent models of chronic pain, but the underlying mechanisms remain unknown. Using two mouse models of neuropathic pain, we show the intrinsic properties of PBN neurons are largely unaltered following injury.

The spared nerve injury model of neuropathic pain differentially altered PBn monosynaptic excitatory input to CeA neurons based on molecular identity and topographical location within the CeA. These results provide insight into the functional organization of affective pain pathways and how they are altered by chronic pain.
