Here, we report that cationic cholesterol-dependent tropism can differ from cationic helper lipid–dependent tropism, along with the development of an LNP that delivers mRNA to the heart as well as lung stem cells. These data provide evidence that charge-dependent tropism may be useful for genetic diseases requiring delivery to multiple tissues.

2023年4月1日 · Our objective herein was to examine LNP delivery of mRNA in in vitro and in vivo scenarios pertinent to pulmonary fibrosis for eventual modulation of therapeutic protein expression. Varying formulations of LNPs were initially tested to determine efficacious design components that would ensure enhanced mRNA transfection.

LNPs have mostly been targeted to specific cell types or organs by physicochemical targeting in which LNP's lipid compositions are adjusted to find mixtures with the desired tropism. Here lung-tropic LNPs are examined, whose organ tropism derives from containing either a cationic or ionizable lipid conferring a positive zeta potential.
