Structures of PYR complexes have been determined for TbPyK and mammalian M1 and M2 isozymes (Table S1). In these complexes PYR is involved in coordination with the Mg 2+ and forms hydrogen bonds with all residues as observed in the PEP complex except Arg50 (which is hydrogen bonded to the phosphate moiety of PEP in 4HYV).

Our results suggest that Ikaros targets two types of chromatin-remodeling factors—activators (SWI/SNF) and repressors (NuRD)—in a single complex (PYR complex) to the β-globin locus in adult erythroid cells.
