Here, we have developed pH-insensitive fluorescent tools to monitor STIM1 activation and examined the impact of pH i changes on STIM1 function. Our findings reveal that, beyond its roles as an ER Ca 2+ sensor, STIM1 can sense fluctuations in cytosolic pH i through two conserved histidine residues.
