A TCF4-dependent gene regulatory network confers ... - Cell Press
2024年1月4日 · Targeting TCF4 genetically or pharmacologically, using a bromodomain inhibitor, increased immunogenicity and sensitivity of MES cells to ICB and targeted therapy. We thereby uncovered a TCF4-dependent regulatory network that orchestrates multiple transcriptional programs and contributes to resistance to both targeted therapy and ICB in melanoma.

NKT Cell-TCR Expression Activates Conventional T Cells in Vivo, …
To address this issue, we developed a mouse strain allowing conditional Vα14i-TCR expression from within the endogenous Tcrα locus. We demonstrate that naïve T cells are activated upon replacement of their endogenous TCR repertoire with Vα14i-restricted TCRs, but they do not differentiate into NKT cells.
How transcription factors drive choice of the T cell fate
2020年9月11日 · Recent evidence has elucidated how multipotent blood progenitors transform their identities in the thymus and undergo commitment to become T cells. Together with environmental signals, a core...

2020年3月18日 · Mouse invariant natural killer T (NKT) cells are a subset of T lymphocytes which have been shown to play a significant role in innate and adaptive immune responses.
The molecular properties of the bHLH TCF4 protein as an …
6 天之前 · Transcription factor 4 (TCF4), also known as immunoglobulin transcription factor 2 (ITF2) or SL3-3 enhancer factor 2 (SEF2), is a member of the class I bHLH (basic helix-loop-helix) family of transcription factors [], called E proteins.Other known representatives of mammalian E proteins are TCF3 and TCF12 [1, 2].TCF4 is expressed in several tissue types, but mainly in the nervous system, where ...
SOX4 controls invariant NKT cell differentiation by tuning TCR ...
Here we show that the High Mobility Group (HMG) transcription factor (TF) SOX4 controls the production of iNKT cells by inducing MicroRNA-181 (Mir181) to enhance TCR signaling and Ca 2+ fluxes in precursors. These results suggest the existence of tailored, permissive gene circuits in iNKT precursors for innate-like T cell development.
The biology of NKT cells - PubMed
NKT cell biology has emerged as a new field of research at the frontier between innate and adaptive immunity, providing a powerful model to study fundamental aspects of the cell and structural biology of glycolipid trafficking, processing, and recognition.
